Research Focus: Characterization of microvesicles released from podocytes

Our group may provide contribution to the European Kidney-Podocyte Research Platform by developing the following tasks:

  • Characterization of extracellular vesiscles (EVs) released from podocytes after stress induction ( stimulation with: cytokines, antibodies against phospholipase A2 Receptor (PLA2R), sera of patients with focal segmental glomerulosclerosis (FSGS), anti-HLA antibodies, anti–neutral endopeptidase antibodies; MAC, C3a, C5a, hypoxia) with the aim to re-create in vitro some specific pathological conditions that have detrimental effects on these cells in vivo;
  • Identification of EVs expressing specific markers in urine of patients with different pathological conditions.

To reach these goals we may use the following technical and clinical available resources:

  • Immortalized cell lines of human podocyte;
  • Instruments and technologies for molecular analysis of protein, mRNA and miRNA  expression pattern of EVs purified from supernatants of cultured cells or biological fluids (Available Instruments: NanoSight LM10 Nanoparticle Analysis System, Zetasizer, Guava® easyCyte™ 8 Cytometer)
  • Patients with different types of glomerulonephritis and kidney transplanted patients of the Torino Dialysis and Kidney Transplantation Unit.
